So does Montalban, but his voice leaves something to be desired, and the chemistry between the two is more formulaic than shot through with the excitement and fear of attraction. Baldwin, with her lovely, pitch-perfect voice and English certitude makes her arguments persuasively throughout. Rona Figueroa and Matthew Uzarraga in “The King and I” (Photo: Todd Rosenberg)Ĭrucial to any production of “The King and I” is the chemistry between Anna and the King. He is the son of Lady Thiang (the warm yet authoritative Rona Figueroa), the wife who clearly loves the King, and will learn “the scientific” thinking of the West. Chief among them is the King’s young heir, Prince Chulalongkorn (Matthew Uzarraga, who has understood how to command a stage since he was a tot, and does so to exceptional effect here). Anna has been hired by the King of Siam (Paolo Montalban) to serve as schoolmistress to some of his 77 children (those born to the favorites among his many wives). The story, of course, is about the arrival in Siam of the attractive, proto-feminist Welsh woman, Anna Leonowens (played by Kate Baldwin), a worldly widow with a young son and some diplomatic connections. Paolo Montalban and Kate Baldwin star in the Lyric Opera of Chicago production of “The King and I.” (Photo: Todd Rosenberg)īased on a true story turned into a 1944 novel by Margaret Landon, “The King and I” arrived on Broadway in 1951, just as this country was embracing its post-World War IIsuper-power status and still dealing with racial segregation. And Blakeley has used a mostly presentational style (the story stops when a major song is to be sung), that is not just antiquated for Broadway, but for opera as well. This production, initially created by director Lee Blakeley for the Theatre du Chatelet in Paris, has been re-cast with a number of Broadway veterans and Chicago actors. But I was not really heartbroken by this extraordinary, ever timely story that deals with everything from cross-cultural connections and misunderstandings, to the tension between maintaining tradition and adapting to the modern world, to the balance of power between men and women. While I remember leaving the 2014 Marriott Theatre production of “The King and I” in tears, I left the Sunday afternoon performance of the Lyric production feeling I’d seen the heft of the royal treasury of the Kingdom of Siam in the 1860s. Use these values for seeding your parameters in the next step. The inflection point on the curve is parameter c and b is the degree of curvature. When you plot the data you can see the top of the curve which equals parameter a and the bottom is parameter d. Plot(stdcrvdata$logconc, stdcrvdata$OD, main='log standard curve', xlab='x=log(conc)', ylab='y=OD') Get your data into a dataframe and take the log of the concentration (R code in red): X is log concentrations, y is OD signal read from your instrument. You can fit an ELISA curve using free software called R. Where is the 4 parameter option in GraphPad Prism? Do i really have to do 4 parameter logistics or linear regression is also fine (the values i get are copmpletely different).Ģ. I can also use SPSS for the data analysis, but i am not sure whether that choice will be available plus i will have to learn to use the program from scratch.ġ. Does it have any other name (synonym) or is there a specific equation that i can fill in in custom analysis? And if there is one, there are also some A,B,C,D values i have to fill in that i don't know anything about. I know it belongs in the non linear category (i think) but its not in the drop down menu of available analysis. Up to now i was using linear regression for getting the values, but according to the manufacturer this is not the correct way to process the data. The data type are simple and straight forward, meaning i get some optical density values from samples of known concentration and from that i interpolate to find the values of samples of unknown concentration (cytokines). I am using GraphPad Prism 4 for the statistics and i can't find anywhere this type of analysis - it simply isn't there. I am doing research ELISA (I am a medical doctor) and the kit manufacturer says on the manual that i should do 4 parameter logistic analysis of the results i get. Dont know mutch about statistics and i thought some of you might be able to help.
